Introduction: Alzheimer's disease (AD) is a progressive neurodegenerative disease. The most important indicators of AD are amyloid plaque deposition and tau hyperphosphorylation. The present study aimed to investigate the effect of linalool and ascorbic acid-chitosan nanoparticles on the deposition of amyloid plaques in the CA3 hippocampus of AD model rats.
Materials and Methods: Forty male Wistar rats were divided into eight groups, including Control, AD, phosphate-buffered saline (PBS), AD-Chitosan, AD-Linalool, AD-nano Linalool, AD-Ascorbic acid, and AD-nano Ascorbic acid groups. The AD model was induced in rats by bilateral intracerebroventricular injection of beta-amyloid (5 µg in 2 µl). Treatment with linalool (25 mg/kg), nanolinalool (25 mg/kg), ascorbic acid (100 mg/kg), and nanoascorbic acid (100 mg/kg) was performed by gavage for 30 days. At the end of the period, after staining the hippocampus with thioflavin S, the amount of amyloid plaque deposition in the CA3 region was examined.
Results: The percentage of amyloid plaque deposition in the CA3 region of the hippocampus of the AD-Linalool, AD-nano Linalool, AD-Ascorbic acid, and AD-nano Ascorbic acid groups was significantly lower compared to the AD group (p<0.001). Plaque deposition was significantly reduced in the AD-nano Linalool group compared to the AD-Linalool group (p<0.05), and in the AD-nano Ascorbic acid group compared to the AD-Ascorbic acid group (p<0.01).
Conclusion: Encapsulation of linalool and ascorbic acid in chitosan nanoparticles increased the ability of these natural compounds to reduce amyloid plaque deposition in the hippocampus of AD model rats. Encapsulation of these compounds may enhance their neuroprotective effect in the AD model rats.
