Introduction: The emergence of multidrug-resistant Pseudomonas aeruginosa requires alternative therapeutic approaches. This study aimed to investigate the effect of Thyme extract on the expression of the MexXY-OprM efflux pump of this bacterium.
Methods: Different concentrations of thyme extract ethanolic solvent were prepared, and the minimum inhibitory concentration (MIC) and minimum Bactericidal concentration were investigated on clinical strains of multidrug resistant Pseudomonas aeruginosa by microdilution method. Expression of mexX gene was evaluated by comparison with standard strain. The purity and concentration of extracted RNA were determined, and cDNA was synthesized with the relevant kit. qRT PCR process was performed with specific primers. Isolates exposed to the extract were grown at a concentration lower than the MIC, and gene expression was examined in presence and absence of the extract.
Results: The MIC values were obtained at 250 and 125 for 7 and 3 isolates, respectively. The MBC results were 125 and 25/31mg/ml. The changes in the mexX gene expression in bacteria were evaluated based on their relative comparison with the standard Pseudomonas aeruginosa PAO1 and together. Of 10 isolates, 6 samples showed a significant decrease in their mexX gene expression (P<0.05). Sample S2 showed an increase in expression. A significant decrease (P<0.05) was indicated in the mexX expression in 6 isolates with multiple resistance, suggesting that thyme extract reduce the activity of efflux pump. Furthermore, 3 samples had a decrease; however, the level of the mexX expression reduction was not significant.
Conclusion: These findings suggest the use of Thyme extract as an adjunctive therapy against multidrug-resistant Pseudomonas aeruginosa, especially targeting efflux pumps. Further research is needed to optimize its formulation.
